OPTIMIZATION OF EXTRACTION TECHNOLOGY OF GENTIOPICROSIDE FROM GENTIANA STRAMINEA MAXIM USING RESPONSE SURFACE METHODOLOGY ON ACCOUNT OF HPLC

Extraction time, ratio of methanol aqueous solution to material weight, and methanol concentration for three factor and three level response surface methodology (RSM) was used to study the optimum extraction technology of gentiopicroside from Gentiana Straminea Maxim. The analysis was performed on a high performance liquid chromatography (HPLC) system with gentiopicroside peak areas as response values. A ternary quadratic polynomial model with good correlation (R-2=0.9786) was constructed and employed to guide the gentiopicroside extraction process. The optimum extraction conditions were an optimized extract time of 79.4min, a ratio of methanol aqueous solution to material weight of 23.7mL/g, methanol concentration of 92.6%, and three times extraction. Under the optimization extraction technology conditions, the predictive value of gentiopicroside peak area of the sample solution was 2120.6, and only 2.03% differ from the mean measured value 2164.6, and the optimized mean extraction yield of gentiopicroside was 78.3mg center dot g(-1). Through solvent distribution method crystallization of the extractive from response surface experiments, the white crystalline compound gentiopicroside was prepared and its purity was greater than 95%.
