Multidimensional multinuclear solution NMR studies of encapsulated macromolecules

摘要
The application of multidimensional multinuclear solution nuclear magnetic resonance (NMR) technique which involve encapsulation of the hydrated macromolecules of interest within a shell of surfactant, was analyzed. To illustrate the approach, recombination human ubiquitin was encapsulated within sodium bis(2-ethylhexyl)sulfosuccinate (AOT) reverse-mocelles prepared in n-heptane. Accurate analysis of sensitivity was measured under the influences of intrumental instabilization and true thermal noise. The combination of cold-probe technology and encapsulation of proteins within reverse micelles dissolved in low viscosity solvents was found to be perfect amalgamation of methods.
