摘要
[Objective] To study Effects of p,p'-DDE on the Intracellular ERK MAPK signal transduction pathway in Rat Sertoli Cells cultured in vitro and the mechanisms of it. [Methods] The Sertoli cells from testicular tissue of rats were exposed to different concentrations of p,p'-DDE. Cellular viability of Sertoli cell, and the changes of intracellular p-ERK were determined by immunohistochemical SABC, MTT chromatometry and Western blotting method respectively. [Results] It showed that intracellular p-ERK increased with the dose of p,p'-DDE and had a dose-dependent trend. Cellular viability decreased with increased dose of p,p'-DDE and had significant difference compared with the DMSO control at the concentration of 70 mu mol/L(P < 0.05). p-ERK of Sertoli cell has significant difference at 12 h and 24 h time point compared with the untreated at 50 mu mol/L(P < 0.05). [Conclusion] p,p'-DDE could result in the phosphorylation of ERK which induces the activation of the downstream pathway to influence the transcription land expression of some genes.
