Comparative studies on the mechanism of action of serotonin in frog, rat and human adrenocortical cells

Serotonin (5-HT) is a potent stimulator of corticosteroid secretion in amphibians and mammals, but the effects of 5-HT appear to be mediated by different receptor subtypes, i.e. 5-HT4 receptors in frog and human, and 5-HT7 receptors in rat. The aim of the present study was to determine the transduction mechanisms associated with activation of the different 5-HT receptor subtypes in frog, rat and human adrenocortical cells. In the three species, 5-HT induced a significant increase in cAMP formation by adrenal fragments. The application of 5-HT in the vicinity of cultured frog, rat or human adrenocortical cells also caused a substantial increase in [Ca2+](i). The involvement of CAMP and calcium rise in the mechanism of action of 5-HT was studied on perifused frog and rat adrenal slices. In both species, the agonist-induced aldosterone secretion was significantly reduced in the presence of PKA inhibitors or in Ca2+-depleted medium. These data demonstrate that, while the types of adrenal serotonergic receptors differ from one species to the other, the mechanism of action of 5-HT on adrenocortical cells has been conserved during evolution.
