Comparison of Methods for Total Microbial DNA Extraction from fermented meat

摘要
DNA was extracted from microorganisms in fermented meat by SDS (sodium dodecyl sulfate) method,lysozyme method, benzyl chloride method,bacteria DNA extraction kit method and improved CTAB(cetyl trimethyl ammonium bromide) method respectively. The quality of the DNA were evaluated with purity and yield of the DNA and RAPD amplification polymorphism. Results showed that there are some differences in purity and yield among those five methods. Both benzyl chloride method and improved CTAB method have better yield, but the purity of the DNA by bacteria DNA extraction kit method is better than that of others. The genome DNA extracted by five methods reaches beyond 23kb in DNA size and good integrity when assayed in gel electrophoresis. The genome DNA extracted through benzyl chloride method is the most clear and sharp figure. In a word, the benzyl chloride method is most suitable for extracting genomic DNA from microorganisms in fermented meat.
