Apoptotic Signalling of Huh7 Cancer Cells by Biofabricated Zinc Oxide Nanoparticles

摘要
The development of drug for cancer is the major task for the researchers in world wide. The liver cancer is one of the primary types of cancer affected by many people all over the world. Zinc oxide nanoparticles place a vital part in the medical research. The present work emphasis on the synthesis of biofabricated Zinc Oxide nanoparticles in an ecofriendly and cost-effective method using Luffa acutangula peel extract. The nanoparticle synthesized was characterized by UV visible absorption spectroscopic studies and showed maximum absorption peak at 377.77 nm corresponding to Zinc oxide nanoparticles. The SAED pattern of biofabricated ZnO nanoparticles shows the crystalline nature and the size of nanoparticles are found to be 18 nm. The biofabricated Zinc oxide Nanoparticles have cytotoxicity against Huh7 liver cancer cells as that of the Doxorubicin drug. The cytotoxic study of biofabricated ZnO nanoparticles by MTT assay against Huh7 liver cancer cell lines showed a dose-dependent effect and the lethal concentration (LC50) of ZnO nanoparticles was confirmed to be 40 mu g/ml. The stimulation of apoptotic signaling pathway in Huh7 cancer cells has been studied by Reactive Oxygen Species (ROS) assay, Rhodamine (Rho123) assay and Hoechst assay. The results of this study concluded that Zinc oxide Nanoparticles synthesized using Luffa acutangula peel shell extract act as a cost effective therapeutic alternative agent for liver cancer treatment.
