Studies on Serum of Arsenic Trioxide Treated Rats by 1H NMR Based Metabonomics

摘要
The metabolites of rats' serum caused by arsenic trioxide intragastric infusion (dosed at 2, 10 mg/kg body weight, respectively) were investigated through metabonomics. The metabolic profiling detected by NMR subjected to principal component analysis (PCA), which find out the different metabolites of the control group and arsenic trioxide treated group, to explore the acute biological effects. The results revealed that arsenic trioxide caused acute damage on rats' liver, and the toxicity effect increased with the increase of dose. Low dosage administration arsenic trioxide gave rise to reactive oxygen species (ROS), and high dosage administration arsenic trioxide induced cell apoptosis. Sugar, amino acid fat metabolism disorder and energy metabolism abnormality occurred in both low and high dosed rats.
