A monoclonal antibody-based lateral flow dipstick for the rapid detection of chlorimuron-ethyl in soil sample

摘要
A monoclonal antibody-based lateral flow dipstick is developed in which the detection reagent is colloidal gold labeled affinity purifed monoclonal anti- chlorimuron-ethyl antibody. The capture reagent is chlorimuron-ethyl-bovine serum albumin conjugate and goat anti-mouse antibody which is immobilized on the nitric cellulose (NC) membrane. The visual detection limit for chlorimuron-ethyl is 100 WL. Aanalysis can be completed in less than 10 min. Eight soil samples were fortified with chlorimuron-ethyl in a concentration range of 0 - 2000 mu g/L and extracted with 0.15 mol/L NaHCO(3), for 2 h at room temperature. The supernatant was collected and analyzed directly with dipstick test and gas chromatography. When the cut-off value was set at 100 mu g/L to distinguish the positive and negative samples, The results from the two methods are completely identical. The monoclonal antibody-based lateral flow dipstick can be employed for rapid and simple screening for the residue of chlorimuron-ethyl in soil samples.
