摘要
An efficient palladium-catalyzed coupling between a pyrazole amide and a heteroaryl bromide was developed to enable the streamlined synthesis of a lead candidate for schizophrenia treatment. Process robustness and scalability were achieved using ethanol as a solvent. The Xantphos ligand employed in the catalytic system was found to be key to prevent common side reactions arising from the alcoholic solvent. The scope of the methodology was investigated and is presented.
