Radioligand Therapy With 177Lu-PSMA for Metastatic Castration-Resistant Prostate Cancer: A Systematic Review and Meta-Analysis

OBJECTIVE. Several clinical studies have shown the efficacy of Lu-177-labeled prostate-specific membrane antigen (PSMA) radioligand therapy (RLT) for metastatic castration-resistant prostate cancer (mCRPC). The purpose of this article is to present results of a systematic review and meta-analysis aimed at compiling and outlining efficacy and safety data on Lu-177-PSMA RLT for mCRPC across all studies published to date. CONCLUSION. The results of the systematic review and meta-analysis suggest that Lu-177-PSMA RLT is an effective treatment of advanced-stage mCRPC that is refractory to standard therapeutic options and that it has a low toxicity profile. High-level evidence from randomized control trials is crucial for confirming the effectiveness of Lu-177-PSMA RLT and for instituting this therapy in the routine clinical care of patients with mCRPC.
