Nonsteroidal anti-inflammatory drugs (NSAIDs) have been shown to be one of the most affective class of chemoprotective agents to date. Most of the published studies have evaluated gastrointestinal malignancies. Here the authors review the clinical, epidemiologic, and laboratory data evaluating the affects of NSAIDs in cancer prevention. The authors hope that through a rigorous evaluation of past work and future experimentation the molecular basis for cancer chemoprevention by these agents will be understood.
