MiRNA-10a-3p ENHANCED CELL APOPTOSIS BY REGULATING PTEN/PI3K/AKT SIGNALING PATHWAY IN BREAST CANCER

摘要
Introduction: Breast cancer is a heterogeneous disease that requires individualized treatment according to different molecular types. Currently, it can be divided into Luminal A type, Luminal type B, Human Epidermal Growth Factor Receptor-2 (HER2) positive type. Early breast cancer often does not have typical symptoms and signs, if the treatment is not timely, it can be life-threatening. Therefore, it is most important to find the efficient methods to cure breast cancer((1-3)). Methods: The target genes of miRNA-10a-3p were predicted by Targetscan, miRWalk, miRsystem databases and the miRNAs which regulate the expression of Phosphate and Tension Homology Deleted on Chromsome Ten (PTEN) was predicted by cytoscape in this study. The MCF10A Cells were served as control group, MCF-7 cells was used as model group. The miRNA inhibitor was transfected into the model group, the relationship between miRNA-10a-3p and PTEN was verified by Luciferase reporter gene, and the expression of miRNA-10a-3p, PTEN and Phosphatidylinositide 3-k-incises (PBK), Protein Kinase B (AKT) mRNA were determined by Reverse transcription Polymer ase Chain Reaction (RT-PCR). The expression of PTEN, PI3K, p-PI3K,AKT and p-AKT proteins were detected by western blot. Results: Our research indicated that 19 downstream target genes regulated by miRNA-10a-3p and PTEN, and the expression was negatively correlated with PTEN by transfecting miRNA-10a-3p inhibitor through the database prediction and the intersection of multiple databases. MiRNA-10a-3p is highly expressed in MCF-7 cells, and it can treat breast cancer by increasing the target gene PTEN expression and regulating the expression of PI3K/AKT signal pathway. Conclusion: MiRNA-10a-3p can be used as a biological marker of breast cancer, which may enhance the cell apoptosis by regulating PTEN/PI3K/AKT signal pathway in breast cancer.
