摘要
Prion diseases are progressive and fatal neurodegenerative disorders in humans and animals and involve conformational conversion of cellular prion proteins (PrPC) into the pathological isoform (PrPSC). The article presents experiments in transgenic animals, tissue culture cells, sequential analysis and NMR of PrPC measurements carried out in order to identify the region in the PrPC important for PrPSC formation. These studies have all revealed that the degree of homology between secondary structures in the middle third PrPC molecule in particular animals' is crucial in destroying the species barrier in prion diseases.
