Effective Agrobacterium-mediated transformation of chicory (Cichorium intybus L.) with Mycobacterium tuberculosis antigene ESAT6

摘要
Conditions of high efficient chicory (Cichorium intybus L.) transformation with Mycobacterium tuberculosis antigene ESAT6 responsible for the maximal frequency of regeneration of green plants with transformed DNA have been determined. Transformation frequency was up to 86% when the cotyledons were cultivated within 3 days without cefotaxime followed by 1 day of cultivation without kanamycine. According to PCR-analysis, the presence of both selective nptII and target esxA genes was shown in all analyzed plants. At the same time, RT-PCR analysis demonstrated the presence of nptII transcripts in eight analyzed lines and esxA transcripts in only five analyzed lines.
