A NOVEL METHOD FOR RELEASE SOMATIC EMBRYOS FROM WHEAT (Triticum aestivum L.)

摘要
In this study, immature embryos were used as an explant. The effect of supplementing different concentrations of 2,4-dichlorophenoxyacetic (2,4 D) combined with 0.5 mg L-1 Kinetin, 2 mgL(-1) of Naphthalene acetic acid (NAA), and 2.9 gL(-1) of casein hydrolysate on somatic embryogenesis production was studied. The highest callus induction percentage was for Cultivar Giza 168 (93.3%) on callus induction media 2 (CM2T). The concentration of 2 mg L(-1)of 2,4-D in combination with 2 mg L-1 NAA, and 2.9 mg L-1 hydrolyzed casein was the optimum condition to improve in vitro somatic embryogenesis. Misr2 was the only regenerative cultivar on regeneration media 1 (RM1) with (25%). The present study indicated that calli transferred to CM2T contained a different concentration of Seawater (25%, 50%, 75%, and 100 %) produced somatic embryos but the embryos did not germinate, So further studies are still required to identify the real reasons for lacking germination of the obtained somatic embryos.
