Monoclonal antibodies against Tobacco rattle virus and its application in the detection of TRV isolates

摘要
Two murine hybridoma cell lines secreting monoclonal antibodies (MAbs) against Tobacco rattle virus (TRV) were produced by fusion of mouse myeloma cells (Sp2/0) with spleen cells from BALB/c mice immunized with purified TRV isolate PpK20. Western blot results indicated that both MAbs could react with the coat proteins of four TRV isolates tested. Triple antibody sandwich enzyme-linked immunosorbent assay (TAS-ELISA) was optimized with produced MAbs. Also, the sensitivity of TAS-ELISA was compared with that of indirect-ELISA. Finally, immunocapture RT-PCR (IC-RT-PCR) was established for reliable and efficient detection of-my. The two established assays could be used to detect rapidly and reliably TRV in the field plants.
