A rapid and easy method to simultaneously determine the concentrations of cell culture process metabolites is the analysis with Near-infrared Spectroscopy. We used a multivariate Partial Least Squares (PLS) algorithm to calculate a model for the prediction of glucose, lactate, glutamine, ammonia, glutamate and human antithrombin III in a production process with recombinant CHO cells. Once calibrated for a given system (medium composition, concentration ranges) with a sufficient number of samples, the pre-treatment, measurement and prediction of these substances from one spectrum takes only 10 minutes.
