Local drug delivery from hydroxyapatite ceramic fibres

Hydroxyapatite (HA) ceramic fibres were fabricated by diffusion-induced phase separation. The fibres were loaded with riboflavin phosphate (RP) as a model drug of water soluble antibiotics to treat orthopedic infections and subsequently spray-coated with polylactide (PLA) or poly(lactide-co-glycolide) (PLGA). An in vitro dissolution test showed a burst release within the first day from fibres with a 3 mu m PLA or PLGA coating. This initial burst release was already reduced by a 7 mu m PLA or PLGA coating. The dissolution profile of fibres with a 14 mu m PLA or PLGA coating showed a RP release of 80% after 28 days. The 14 mu m PLA coating exhibited a lag phase of two days, followed by a near zero order release until the 21st day.
