Dot blot assay coupled with optical detection was developed to determine Salmonella typhi (S. typhi) to obtain the low-cost diagnosis. S. typhi was first dotted on nitrocellulose membrane, and then captured by polyclonal rabbit antibodies to S. typhi 0901 labeled with colloidal gold. The wine red dots appeared on nitrocellulose membrane and the results could be evaluated by naked eyes. For optical detection, the strips were captured by scanner and AdobePhotoshop was used to monitor the mean density of each dot. The limit of detection (LOD) was found to be 0.14 mg/mL of S. typhi which could be evaluated by naked eyes. The linear region of optical detection was observed with a good coefficient of 0.9986.
