The effect of ANGPTL8 protein on proliferation and apoptosis in HepG2 hepatocellular carcinoma cell line

摘要
Hepatocellular Carcinoma is that the 6th most generally analyzed malignancy worldwide and also the helpless visualization of the disorder; it's the fourth driving reason for death within the world. The ANGPTL protein family has seven typical members (1-7); and one atypical member, ANGPTL8 (Angiopoietin like Protein 8). It's discharged in the liver, white and brown adipose tissue. ANGPTLs assume a significant part in glucose metabolism, lipids, hematopoiesis, cancer, and inflammation. ANGPTL8 affects lipid metabolism and fundamentally raised lipogenesis in HCC cells. According to studies conducted in the HepG2 cell line, ANGPTL8 increased cleavage and discharge of the functional ANGPTL3. In HCC, ANGPTL3 forestalled cell expansion through the downregulation of p38MAPK and MMP-9 cascade's activation. HepG2 cells were treated with different doses of ANGPTL8, and the mRNA expression of BAX and BCL2 genes was evaluated by q real-time PCR. Flow cytometry investigation was wont to the analysis of treated cells apoptosis. Used annexin V binding to PS and fluorescent tracker of Annexin V for PS identification on the outer layer of apoptotic cells. BCL2 gene is an anti-apoptotic, and also the BAX gene is apoptotic. ANGPTL8 diminish BCL2 gene expression and increment the expression of BAX gene. It assumed that ANGPTLs play an essential role in tumor cells development. The field of research about ANGPTL proteins may need new concentrate on the advancement of future investigation.
