摘要：

We designed and expressed a novel mutant MspA in E. coli by replacing three negatively charged residues in the constriction zone with neutral residues and adding three arginine substitutions around the constriction zone. Our results showed that the Mut-MspA had good channel-formation ability and also slowed down the ssDNA translocation rate ten times, which could facilitate its applications on single molecule detection and characterization of DNA.
