OXIDATIVE STRESS-INDUCED ALTERATIONS IN FIBROBLAST PLASMA MEMBRANE LIPIDS

摘要
The alterations in the lipid composition and the raft microdomain content in plasma membranes of fibroblasts exposed to oxidative attack have been investigated. The results show that treatment of cells with hydrogen peroxide induces reduction of the raft microdomain lipid components, sphingomyelin and cholesterol, as well as of some of the most unsaturated phospholipids phosphatidylserine and phosphatidylethanolamine. The eventual protective role of sphingomyelin against oxidative damage of plasma membrane lipids has been tested by treatment of cells with sphingomyelinase, which selectively hydrolyzes membrane sphingomyelin. An inversely proportional correlation has been established between the level of lipid peroxides and the relative content of raft microdomains.
