Determination of the monoamine neurotransmitters in rat striatum by microdialysis sampling coupled with high performance liquid chromatography-electrochemistry detection

摘要
Microdialysis sampling coupled with high performance liquid chromatography and electrochemical detector was developed and applied to determine the monoamine neurotransmitters in rat striatum. The results indicate that the peak currents of dopamine (DA), 3, 4-dihydroxyphenyl acetic acid (DOPAC) and 5-hydroxylindole acetic acid (5-HIAA) are linear to their concentrations ranging from 3.0 X 10(-8) to 1.0 x 10(-5) mol/L with the calculated detection limits of 1.2 x 10(-8) mol/L for DA, 1.5 X 10(-8) mol/L for DOPAC and 2.0 x 10(-8) mol/L for 5-HIAA, respectively. In order the study the physiological function of nitric oxide on striatal DA release, a continual perfusion of 1.0 x 10(-5) mol/L sodium nitroprusside was performed into rat striatum, which resulted in a 50% increase in DA level.
