Proteomic Analysis of Pine Nut (Pinus Koraiensis Sieb. et Zucc) Protein in Changbai Mountain by Shotgun Approach

摘要
Protein of Pinus koraiensis Sieb. et Zucc were identified by shotgun proteomics, 827 proteins were obtained. Using CD-HIT method to remove protein with sequence similarity over 95%, 802 proteins were left for Gene Ontology. 441 proteins (54.99%) were in the range of pI 4.00-7.00, which in conformity with the fact that the solubility of pine nut protein reached the minimum when pH 4.5. 76.81% proteins were mainly distributing in a range of 10-50 kDa; The protein numbers of residues ranged from 100 to 200 was 219; 85.16% proteins were hydrophilic and hydrophobic proteins accounted for only 14.84%; Pine nut proteins could be divided into 15 categories by biological process, of which 561 proteins related to metabolic process. Proteins were classified by molecular function, of which 396 proteins had catalytic activities, 356 proteins with binding activity. According to the cellular component, 398 proteins were from the cell and 288 proteins from the organelles. Shotgun strategy provided reliable theory basis for the development of edible and medical value of pine nut protein, which also had important reference value for the research of wild resource in Changbai mountain of Jilin province.
