The combination of metal Zn(II) ions with a pyrazine functionalized linker 2,3,5,6-tetrakis(4-carboxyphenyl)pyrazine (H4tcpp) affords a new nontoxic porous Zn(II)-based metal-organic framework (MOF) [Zn-3(OH)(2)(FlztccP)(2)(bPY)(2)](H2O)(3)(DMF)(3) via a hydrothermal reaction with the presence of the assistant ligand 4,4'-bipyridine (bpy). Furthermore, we can acquire the nanostructure 1 through utilizing green grinding approach. Nitrogen adsorption measurements showed the existence of micropores along with a high BET surface area that in activated nanostructure 1 (1a). We can see from the drug loading experiment that 5-fluorouracil (5-Fu) is captured into the nanostructure 1a's pore preferentially with a loading capacity of 30.7%. At the same time, the 5-fluorouracil was controlled to release in the artificial human body with a phosphate buffered saline solution. The anti-viability and anti-proliferation activity of 5-Fu@1a on CoC1 ovarian cancer cells was carried out by Cell Counting Kit-8 (CCK-8) assay and clone formation assay, the data forecasted 5-Fu@1a has excellent anti-cancer effect in vitro. To explore the manner of cancer cell death, the apoptosis assay and Western blot was carried out, and the results suggested in a caspase-dependent manner, 5-Fu@1a can attract CoC1 ovarian cancer apoptotic death. The remarkable inhibitory activity in mice of 5-Fu@1a has been further confirmed via the in vivo results of tumor volume and body weight. (C) 2019 Elsevier B.V. All rights reserved.
