Leptospires have been isolated from the testes of cattle (Ellis et al., 1986) and from bull semen (Masri et al., 1997). Direct diagnostic methods for isolation of Leptospira organisms from semen are not easily performed due to the rapid growth of other microorganisms. Among the DNA-based techniques, PCR assays have been. applied to cerebrospinal fluids, milk and semen (Veloso et al., 2000). The sensitivity of detecting spiked leptospires in bovine semen sample using three different DNA extraction methods is reported.
