More detailed flow cytometry analysis of lymphocyte subpopulations in common variable immunodeficiency (CVID)

摘要
Flow cytometry analysis of CVID patients lymphocytes showed consistently CD4+ or CD4+45RA+ decline. There are also suggestions from functional studies demonstrating partial defect of early phase of T cell activation. Objective: Perform more detailed immunophenotyping (IPT) of CD4 and CD8 subsets and try to test functional activity of CVID patients lymphocytes. Results and conclusions: I. There was found significantly higher proportion of "Th1-like" (CD4+45RO+62L-) and "Th2-like" (CD4+45RO+62L+) memory phenotypes in CVID, for Th1 also documented by cytoplasmic IFN gamma expression. 2. In Vitro lymphocyte activation (expression of CD69 and CD25 was measured): a) CVID patients showed higher spontaneous activation of CD4+ cells in comparison with both other groups (IgAD, healthy control), b) in case of anti-CD2/CD2R stimulation no activation defect was approved, c) suboptimum phytohemagglutinin (PHA) concentration (2,5 mug/ml) showed a marked decline of activation for CD4+ cells.
