A spectroscopic tool based on an interference filter and birefringent prisms: demonstration of detection of 5-aminolaevulinic acid-induced protoporphyrin IX fluorescence

We describe a differential fluorescence detection system that consists of a 405 nm diode laser for fluorescence excitation, a bifurcated fibre for delivery of light to and collection of light from tissue, birefringent prisms and a tilted filter. This instrument is optimized for the selective detection of protoporphyrin IX fluorescence and autofluorescence from tissue, and its effectiveness demonstrated using low excitation light levels (0.7 mW cm(-2)).
