摘要
Although vaccines against influenza A virus are the most effective method by which to combat infection, it is clear that their production needs to be accelerated and their efficacy improved. A panel of recombinant live attenuated human influenza A vaccines (LAIVs), including NS1-73, NS1-126, NSD5, were generated by rationally engineering mutations directly into the genome of a pandemic-H1N1 virus. The vaccine potential of each LAIV was determined through analysis of attenuation, immunogenicity, and their ability to protect mice and ferrets. The data indicate that the novel NSD5-LAIV was ideally attenuated and elicited strong protective immunity. This study also shows that attenuating mutations can be rapidly engineered into the genomes of emerging/circulating influenza A viruses in order to produce LAIVs.
