Estrogens are carcinogenic causing uterine and breast cancers. The mechanism is unknown. We found that 17 beta-estradiol (E-2) could be activated by the epoxide-forming oxidant, dimethyldioxirane (DMDO). Since epoxidation is required for the activation of many chemical carcinogens, we proposed that epoxidation is the underlying mechanism for the initiation of estrogen carcinogenesis (Carcinogenesis 17,1957-1961,1996). Initiation requires the binding of a carcinogen to DNA forming DNA adducts. In the support of our hypothesis, evidence is presented to show the detection by P-32-postlabeling the formation of E-2-DNA adducts both in vitro after E-2 activation and in vivo in the mammary glands of the female ACI rats after either multiple intramammillary injections of E-2 or a single injection of the DMDO activated E-2.
