Differential expression of TGF-beta 1 and TGF-beta 3 during fibrogenesis in chronic rejection of human liver allografts

In chronic inflammatory livers as well as in chronic rejected liver allografts increased production of extracellular matrix molecules is observed in accordance with macrophage infiltration and lipocyte transformation. While TGF-beta 3 seems to be involved in an autocrine stimulation of lipocyte proliferation in beginning fibrogenesis, apparently macrophage-derived TGF-beta 1 appears to take part in lipocyte transformation and maintenance of fibrotic deposit in later stages of fibrogenesis. The correlation of observations in chronic inflammation and chronic rejection indicates similar fibrogenic mechanisms despite different triggering (viral infection, autoimmune disease, alloreaction).
