Synthesis and pharmacological evaluation of some 2,4-disubstituted hexahydroquinazolines

A series of 2-substituted-4-(p-chlorophenyl)-3,4,5,6,7,8- hexahydroquinazolines (I-VIII) were synthesized by the nucleophilic substitution of 2-bromo-4-(p-chlorophenyl)3,4,5.6,7,8-hexahydroquinazoline 5 with different amines. Spectral data (IR, NMR, and mass spectra confirmed the structures of the synthesized compounds. The purity of these compounds was ascertained by microanalysis. The synthesized compounds were tested for their analgesic, anti inflammatory and antimicrobial activities. While all the test compounds exhibited good analgesic activity, the compounds I and V exhibited comparable activity with the reference standard aspirin. The compound VII exhibited comparable antiinflammatory activity to the reference standard ibuprofen. The test compounds exhibited moderate antibacterial and mild antifungal activity.
