Advantages of Two-photon Microscopy with Ultrashort Pulses

摘要
Ultrashort pulses are expected to be beneficial for multiphoton microscopy. To utilize their advantages, however, chromatic dispersion must be compensated. We use multiphoton intrapulse interference phase scan (MIIPS) to measure and then eliminate phase distortions of pulses with a FWHM spectral bandwidth greater than 100 nm. Once compensated, the transform limited pulses (<15 fs) deliver higher two-photon excited fluorescence signal intensity, which translates into deeper optical penetration depth.
