IL6-174G>C and TNFA-308G>A polymorphisms in Bulgarian patients with COPD and Bronchial asthma

COPD and Bronchial asthma are multifactorial chronic lung diseases driven by abnormal inflammatory reactions. Therefore, inflammatory mediators are considered of distinct importance in pathogenesis of these conditions. In the current case-control studies we investigated promoter polymorphisms in the genes of interleukin 6 (IL6 -174G>C) and tumor necrosis factor alpha (TNFA -308G>A), known to affect the transcriptional levels of the encoded cytokines. We genotyped 164 patients with COPD, 47 with Bronchial asthma and 174 control individuals using PCR-RFLP-based methods. Results: The analyses showed no significant difference in -174G>C IL6 genotype and allele distribution between controls and patients, either with COPD nor asthma. When analyzing -308G>A TNFA genotype and allele distribution, we did not find significant difference between controls and asthma patients. However the minor A allele, as well as A allele carrier status (AA+AG) were less frequent among patients with COPD compared to the controls: 0.058 vs. 0.105 for A allele, p=0.027, and 0.110 vs. 0.192 for GA+AA genotypes, p=0.036. Thus, the AG+AA genotypes turned out to determine about 2-fold lower risk for developing COPD (OR=0.519, 95% CI of 0.279-0.965).
