摘要
An overview of the various ways in which phage display have been applied in the last five years is given. Focus is on the use of the filamentous bacteriophage M13 and its close derivative fd. Techniques for the production of DNA libraries are also covered, specifically, Kunkel mutagenesis and the ligation of polymerase chain reaction (PCR) products into a vector. Furthermore, ten applications of phage display are introduced including: i) affinity reagents; ii) inhibitors; iii) mirror image phage display; iv) binding to inorganic materials and nanostructures; v) selections for improved protein stability; vi) rapid analysis of mutant affinities; vii) artificial transription factors; viii) identification of protease substrates; ix) enzyme design; and x) cell targeting.
