Cytokines (TGF-β, MIP-1α) and nitrogen oxide level in induced sputum from patients with COPD or asthma, and bacterial colonisation of respiratory tract

Aim of the study was to evaluate the immune response to airway bacterial colonisation by measuring the level of MIP-1 alpha, TGF-beta and nitrogen oxide (NO.) in patients with chronic obstructive pulmonary disease (COPD) and bronchial asthma. The study group consisted of 36 patients in a stable state of the disease without any clinical sings of respiratory tract infection. The sputum was obtained by means of sputum induction method. MIP-1 alpha and TGF-beta were measured by ELISA method and NO. by Griess reaction. The sediments were quantitively cultured and cytospines were tested for bacterial antigens by immunofluorescence method. Bacterial colonisation was observed more frequently in patients with asthma (58%) than with COPD (31%). TGF-beta and NO. sputum concentration were markedly elevated in COPID patients with bacterial colonisation in comparison with COPD patients without colonisation. Conversely NO. was reduced in colonised asthmatic patients as compared with the subgroup without colonisation.
