The O-specific polysaccharide (OPS) obtained by mild-acid degradation of the lipopolysaccharide from Aeromonas sobria strain Pt312 was studied by sugar and methylation analyses along with H-1 and C-13 NMR spectroscopy, including 2D H-1, H-1 COSY, TOCSY, NOESY, H-1-detected H-1, C-13 HSQC, and HMBC experiments. The sequence of the sugar residues was determined using H-1, H-1 NOESY and H-1, C-13 HMBC experiments. It was found that the OPS was built up of disaccharide repeating units composed of GlcpNAc and non-stoichiometrically O-acetylated Rhap residues, and had the structure. -> 4)-alpha-L-Rhap-(1 -> 3)-beta-D-GlcpNAc-(1 -> 2/3 | similar to 80/35% OAc (C) 2014 Elsevier Ltd.
