We have used the U.S.E. (Unique Site Elimination) Mutagenesis Kit to introduce mutations into the cDNA encoding the 5-hydroxytryptamine(3) receptor (5-HT3-R) that was cloned into pBluescript*. We found that when working with the U.S.E. Kit and pBluescript, modifications to the recommended protocol were required to achieve optimal results. These modifications are presented, as well as suggestions for primer design and a screening method that we found to be beneficial.
