Effect of hyperthermia on the action of methotrexate against the growth of Ehrlich ascites carcinoma cells

Hyperthermia (HPT) at 43 degrees C for 30 min increased the cytotoxicity activity of the antimetabolite methotrexate (MXT) against the growth of Ehrlich ascites carcinoma cells. There was significant delay in tumour growth with 92% inhibition of tumour volume. There were 74, 29.5 and 36% decreases respectively in the rate of DNA, RNA and protein synthesis of tumour cells after treatment with combination of MTX and HPT. Moreover, combination therapy produced pronounced inhibitory effects on cholesterol, total lipid and acid phosphatase activity. Our results suggest the benefit of using HPT to sensitise tumour cells to the cytotoxic activity of MTX. At the same time, it may sensitise normal tissue to the action of antimetabolite drugs.
