A kinetic analysis is made of an enzymatic reaction with slow-binding inhibition in which the enzyme-substrate complex suffers an irreversible inactivation. The explicite equation for the product of reaction is obtained by solving the system of differential equations describing the kinetics of the enzymatic system, under conditions of enzyme limiting and assuming an initial steady state and a concentration of inhibitor higher than that of the free enzyme. The analytical equations derived have been tested by numerical calculus. A kinetic data analysis is suggested, which leads to the evaluation of the principal kinetic parameters of the reaction. This analysis is based in the progress curve of the product of the reaction. We demonstrate the goodness of the procedure suggested by computer simulation and illustrate it with a numerical example to. (c) 2005 Elsevier Inc. All rights reserved.
