Difference Gel Electrophoresis Based Differential Proteomic Analysis of Hepatocellular Carcinoma Cell Lines with Different Metastasis Potential

摘要
Differential proteomics with difference gel electrophoresis (DIGE) and mass spectrometry (MS) was carried out on the four hepatocellular carcinoma (HCC) cell lines with different metastasis potential to identify HCC metastasis related proteins. Total of 1700 protein spots were found in the gel, 167 of which had quantitative differences between cell lines. Through MS identification, total of 146 differentially expressed protein spots, corresponding to 96 non-redundant proteins were found out. Bioinformatic analysis showed that most metastasis related proteins was involved in cell assembly and movement. Hydroxy-prostaglandin dehydrogenase (HPGD) was found to be positively related to HCC metastasis through inhibiting apoptosis and promoting invasion.
