Determination of phenol and catechol in tobacco leaf by solid phase extraction and high performance liquid chromatography

摘要
A method for the determination of phenol and catechol in tobacco leaf by high performance liquid chromatography with methanol and 0.05 mol/L of monopotassium phosphate as mobile phase at a flow-rate of 0.5 mL/min was described. Phenol and catechol were extracted from tobacco sample by distillation and solid phase extraction with Sep-Park-C-18 cartridge. The analytical column is Nova-Pak-C-18 (3.9 x 150 nun). The detection wavelengths of phenol and catechol are UV-270.3 nm, and UV-275.0 nm respectively. The recoveries of phenol and catechol. are 91.8% similar to 104.5% and 89.6% similar to 103.8% respectively. This method has been applied to the determination of phenol and catechol in tobacco with satisfactory result.
