Cartilage has the limited capability of regeneration. Thus, there is an imperative need to develop new strategies for cartilage tissue engineering amenable for clinical use. Chondrocytes based tissue engineering has exerted a promising approach for cartilage regeneration. However, the occurrence of de-differentiated chondrocytes severely inhibits the successful application of cartilage repair in clinical practice. The thorough comprehension of related factors (drugs, extract of some plants, chemical substances and new synthesized organics) involved in the dedifferentiation is necessary to keep the normal phenotype of chondrocytes for the application of cartilage tissue engineering. This review discusses the current knowledge on the related factors in the context of chondrocyte dedifferentiation, and further presents the current substances that have been utilized to induce or inhibit dedifferentiation, which may direct the improvement of clinical cartilage repair.
