OLODATEROL HYDROCHLORIDE β2-Adrenoceptor Agonist Treatment of COPD Treatment of Asthma

Olodaterol hydrochloride (formerly known as BI-1744-CL) is a novel, once-daily, long-acting beta(2)-adrenoceptor agonist. Its pharmacological profile has been extensively studied during the preclinical stage of the development process. Olodaterol hydrochloride is an enantiomeric pure, selective and potent agonist of the human beta(2)-adrenoceptor. It combines a novel efficacy profile toward the different beta-adrenoceptors, exerting almost full intrinsic activity at the beta(2)-adrenoceptor and weak partial agonism at the beta(1)-adrenoceptor. It has a rapid onset of action and a long duration of action, allowing once-daily administration. Olodaterol hydrochloride shows an improved systemic pharmacodynamic effect profile. Clinical studies have confirmed the 24-hour bronchodilator efficacy and once-daily dosing of olodaterol hydrochloride both in asthma and chronic obstructive pulmonary disease. Treatment with olodaterol hydrochloride was generally well tolerated. Combination therapy with olodaterol + tiotropium appears to provide additional benefits due to their complementary modes of action.
