Effects of low-intensity pulsed ultrasound on proliferation of bone mesenchymal stem cells

摘要
Objective: To explore the feasibility of promoting bone mesenchymal stem cells (BMSCs) proliferation by low intensity pulsed ultrasound(LIPUS). Methods: The third generation of BMSCs was seeded in 6-well plates and randomly divided into 2 groups: LIPUS groups and blank-control groups. Growth of cells was observed daily under light microscope and the cell proliferation was determined by MTT at 0 day pre-irradiation and 1, 3, 5day post-irradiation; Cell cycle changes were analyzed at 0 day pre-irradiation and 1, 3, 5, 7day post-irradiation using the flow cytometry. Results: Cell proliferation of the LIPUS group was significantly more active than that of the control-group. LIPUS group also had more intensive cell colonies; MTT test showed that cell proliferation in the LIPUS group of 1d, 3d and 5d post-irradiation were significantly higher than that in the control group (P<0.05); Cell cycle changes tested by flow cytometry showed that the cell proliferation rate (S+G2%) of the LIPUS-group was significantly higher than that of the control group after irradiation (P<0.01).Conclusion: LIPUS could promote the proliferation of BMSCs and motivate quiescent state BMSCs to enter division state.
