Research on degradation of cellulose using enzyme treatment in flue-cured tobacco by 13C NMR spectroscopy

摘要
Cellulase (CE) and polygalacturonase (PG) were used to treat flue-cured tobacco, and then the content and structure of cellulose in tobacco were determined by C-13 NMR spectra. Neutral aroma substances in tobacco were also determined by the GC-MS method. The results showed that enzyme treatment could decompose cellulose and other cell wall substances in tobacco effectively, and the highest decrease degree for cellulose was 13.5 %. The crystalline allomorph analysis showed that enzyme treatment would lead to a decrease in the ratio of I-alpha and increase in I-beta; however, the paracrystalline ratio increased obviously after CE treatment but decreased after PG treatment. There was a significant increase in the amount of total neutral aroma substances, which increased by 38.7 % at a CE concentration of 20 g l(-1) compared to the control, while the PG treatment showed as much as that of the control.
