Effect of sephadex filtration of buffalo bull semen on freezability and sperm penetration into homologous cervical mucus

摘要
Semen of 4 healthy fertile buffalo bulls (48 collections) was frozen in tris-egg yolk glycerol extender before and after filtration through sephadex G 75-120 to investigate the effect of filtration on freezability and sperm penetration into homologous cervical mucus. Cervical mucus of 10 normal cycling buffaloes in standing heat was pooled to remove the effect of mucus quality on sperm penetration. Filtration of semen through sephadex column resulted in significantly (P<0.05) better maintenance of sperm motility, live sperm count, intact acrosomes and sperm morphology. Sephadex filtered frozen semen showed significantly better sperm penetration into homologous cervical mucus compared to unfiltered semen. It is, thus, concluded that filtration of semen through sephadex column improves the quality of frozen semen.
