A protocol has been developed for the clonal propagation and Agrobacterium-mediated transformation of the Lilium oriental hybrid 'Star Gazer'. Transient GUS expression in basal plate disc-derived tissues and regenerated shoots was achieved following transformation using A. tumefaciens strain AGL1 containing pBI121, the latter carrying the beta-glucuronidase (gus) and neomycin phosphotransferase II (nptII) genes. A mean transformation efficiency of 20% was achieved in three independent transformation experiments. Stable transgene integration and expression were confirmed by PCR and RT-PCR analyses. The efficiency, reproducibility and rapidity of this protocol make it suitable for commercial application and for future research on this important ornamental plant.
