A simple method for the quantitative determination of biogenic amines in wine is described. The diluted sample is passed through an cation-exchange-extraction column to enrich the amines. At the same time amino acids and other substances are eliminated which can cause interference in the following analysis. The separation of the derivatized amines using ortho-phthaldialdehyde was achieved by reversed phase HPLC. For that purpose a new solvent gradient containing phosphat buffer and methanol was developed. Comparing measurements show that the reliability and sensivity of the chromatographic analysis increase considerably after solid phase extraction.
