Thermoresponsive affinity interaction between cells and immobilized antibodies on poly(N-isopropylacrylamide)-grafted surfaces

Anti-human CD90 antibody-immobilized thermo responsive cell culture surfaces were prepared as a model for developing next-generation thermo responsive cell culture dishes. Thymic carcinoma cells (Ty-82) didn't adhere on isotype antibody-immobilized surfaces, and the cells aggregated. By contrast, Ty-82 cells selectively adhere on anti-human CD90 antibody-immobilized thermo responsive surfaces. This result indicated that Ty-82 adhered onto the CD90 antibody-immobilized surfaces through affinity interaction, not through nonspecific interactions. The cells were detached from the surfaces by lowering temperature to 20 degrees C with pipetting. Neonatal normal human dermal fibroblasts (NHDFs) were also adhered on the surfaces, and formed the monolayer. By reducing the temperature to 20 degrees C, NHDFs were spontaneously detached as a single cell sheet. In conclusion, antibody-immobilized thermoresponsive cell culture surfaces for regulating selective cell adhesion were successfully prepared. This surface technology would be useful for selective cell attachment/detachment system such as cell separation.
